Does employing a Spanish person help my autónomo or residency application?​

Potentially, but it is not generally a requirement. Spain's self-employed residence process focuses primarily on the viability of the business, the applicant's qualifications, financial resources, and sustainability of the proposed activity.

The Spanish Government's Start Up in Spain guide states that there are no minimum capital requirements and no minimum number of jobs that must be created. Applications are evaluated on a case-by-case basis.

Hiring Spanish employees may be a positive factor, but is not a requirement or a guaranteed path to approval. The quality and viability of the business itself appear to be the primary factors.
